Tulane vs. Duke Prediction, Betting Pick, Latest CFB Odds for Saturday, September 5th

Tulane and Duke meet in the only Week 1 matchup between reigning conference champions after both programs produced memorable 2025 seasons. Duke enters as the home favorite, but Tulane already beat the Blue Devils last year and remains one of the most respected underdogs on Saturday’s betting board. Check out this Tulane vs. Duke prediction and best bet for Saturday, September 5.

Tulane vs. Duke Prediction: Can Walker Eget Replace Mensah as Transfer QB for Blue Devils?

Tulane travels to Durham on Saturday afternoon to face defending ACC champion Duke at Wallace Wade Stadium. Kickoff is scheduled for 3:30 PM ET on ACC Network, with the Blue Devils favored by 7.5 points in a rematch of Tulane’s 34-27 win last September.

Both programs lost major pieces from championship teams. Tulane begins the Will Hall era after reaching the College Football Playoff, while Duke replaces Miami transfer Darian Mensah with former San Jose State quarterback Walker Eget.

Duke went 9-5 overall and 6-2 in ACC play, capturing its first outright conference championship since 1962 with a 27-20 overtime victory over Virginia. The Blue Devils then closed the season with a 42-39 win over Arizona State in the Sun Bowl, completing a four-game winning streak.

Tulane vs. Duke Line Movement and Betting Splits

Duke opened around -9.5 before the line moved through eight to -7.5. Tulane has received only 53% of spread bets but a massive 79% of the handle, while Duke has 47% of tickets and just 21% of the money.

That is one of the clearest ticket-to-money discrepancies on the afternoon slate. The market is not simply taking Tulane because of last year’s result, with larger wagers helping push the Green Wave through multiple points from the opener.

Tulane vs. Duke Prediction

Tulane is undergoing substantial change under Hall, particularly along both lines of scrimmage, but the Green Wave did not lose everything from their championship roster. Zeon Chriss-Gremillion takes over at quarterback with a skill group that includes Jamauri McClure, Florida State transfer Jaylin Lucas, Anthony Brown-Stephens and LSU transfer Destyn Hill.

That gives Tulane enough speed to challenge a Duke defense replacing all four starters across the defensive line. The Blue Devils have experienced linebackers behind that group, but opening against a mobile quarterback and a deep collection of backs and receivers is a difficult first test for a reconstructed front.

Tulane also retains important experience at the second and third levels defensively. Safety Jack Tchienchou and linebacker Chris Rodgers headline the group, giving Hall established pieces against a Duke offense that is also introducing a new quarterback.

Eget has legitimate experience after transferring from San Jose State, but he is returning from a knee injury and stepping into an offense that previously ran through Mensah. Duke still has championship-level infrastructure and home-field advantage, but replacing the player at the center of the offense creates some uncertainty in the first game.

The matchup from last season should also provide confidence for Tulane. The Green Wave did not steal a fluky game with one late play. They led by double digits at halftime and scored 34 points against the eventual ACC champion.

Duke deserves to be favored because it returns more coaching continuity and plays at home. The problem is the number. Asking the Blue Devils to win by more than a touchdown against another reigning conference champion is a substantial requirement, particularly with Tulane getting the hook above seven.

Tulane vs. Duke Best Bet

Tulane has enough turnover to make an outright repeat of last season uncertain, but +7.5 provides protection in a matchup that was competitive even before both programs changed quarterbacks and personnel.

The betting action strengthens the case. Tulane has 79% of the spread money on only 53% of bets, and Duke has already fallen from around -9.5 to -7.5. With the Green Wave returning enough playmakers to threaten a rebuilt Duke defensive front, taking more than a touchdown is the better side.
