The first matchup was in Week 1 when the Jaguars earned a 31-21 victory over the Colts.
Anthony Richardson started for the Colts in that game. However, it'll be Gardner Minshew getting the ball for the Colts moving forward, thanks to a shoulder injury that Richardson suffered last weekend.
Will Minshew make a difference for the Colts in the second matchup between the two rivals?

Indianapolis Colts vs. Jacksonville Jaguars Prediction
The Colts will play without Anthony Richardson for at least four games. He's out with a shoulder injury, leaving Gardner Minshew as the starting quarterback.
Most teams would be ruined without their starting quarterback. But not the Colts. They've got one of the better backups in the league.
Minshew led the Colts to a 23-16 win over the Titans last weekend. The Colts have averaged over 350 yards of offense, including 226 yards in the air. That number will likely increase with Minshew throwing the ball.
Indianapolis will also get more touches for Jonathan Taylor as he returns from an injury. He signed a three-year extension, so he's the guy, despite Zack Moss' massive game against Tennessee.
However, the depth at running back is something that the Colts have now.
On the other hand, Jaguars have allowed only 81.6 yards per game on the ground. But that doesn't paint the whole picture. The Jaguars aren't great defensively against the run. If the Colts attack the Jaguars in the ground game, they'll have success offensively.
Meanwhile, the Jaguars continue to get good play out of Trevor Lawrence. The team has added over 250 yards in the air per game. The third-year quarterback now has 1,258 yards with five touchdowns and only two interceptions. He's also got the best wide receiver core he's played with in the NFL this season.
But the offensive line hasn't helped the situation at all. The Colts have a legitimate pass rush that will get to Lawrence plenty of times in this game. If Jacksonville's offense line improves, Lawrence will have plenty of options, with Indianapolis struggling in the secondary.
The Colts have also missed countless tackles this season.
Both defenses are very average. The Colts are more dynamic with Taylor in the backfield and should create more big plays down the field using Minshew's arm.
On the other hand, the Jaguars will pick apart Indianapolis' secondary in the passing game.
Therefore, I'll back the Over.
